Tối ưu hóa phản ứng rt-lamp phát hiện nhanh norovirus trong thực phẩm

Norovirus là vi rút thuộc họ Caliciviridae, gây bệnh viêm dạ dày ruột ở người, đối tượng thực phẩm có nguy cơ nhiễm Norovirus thường là các loại nhuyễn thể hai mảnh vỏ và các loại rau quả ăn sống. Trong các phương pháp xác định Norovirus trong thực phẩm, phương pháp RT-LAMP là kĩ thuật khuếch đại đẳng nhiệt cho phép phát hiện nhanh, chính xác với giá thành hợp lý. Tuy nhiên, các bộ mồi LAMP tính đến thời điểm hiện tại chưa đảm bảo độ bao phủ cao cho toàn bộ các kiểu gen của Norovirus. Do đó, nhóm nghiên cứu đã tiến hành nghiên cứu hiệu chỉnh bộ mồi và tối ưu hóa phương pháp RT-LAMP xác định Norovirus trong thực phẩm. Để thực hiện mục tiêu này, các mẫu thực phẩm thu thập trên địa bàn các chợ tại Hà Nội được xử lý, sau đó tách chiết và tinh sạch ARN virus, phản ứng RT-LAMP được khảo sát để chọn ra điều kiện tối ưu (lượng enzyme phiên mã ngược 2U/phản ứng; nhiệt độ ủ 63°C; nồng độ Betain đối với GI: 1M, đối với GII: 0,8M; thời gian ủ: 60 phút) với độ nhạy là 10 phiên bản/phản ứng và độ đặc hiệu đạt 100% trên các chủng khảo sát.

Abstract

Norovirus, a virus of the Caliciviridae family, is the leading agent of food borne viral gastroenteritis worldwide. The most common foods contaminated by Norovirus are bivalve molluscan and raw vegetables. Among methods to detect Norovirus, RT-LAMP method is one of the most efficient low-costed isothermal amplifying techniques that provide rapid and exact specification. However the LAMP primer set does not always have sufficient specificity to detect all the various type of Norovirus. Therefore this study was conducted to optimize the LAMP primer set and RT-LAMP method. With the purpose of accomplishing this goal, the food samples which were collected from some markets in Hanoi were processed, followed by extraction and purification of viral RNA, then the RT-LAMP reaction was tested to select optimal conditions (concentration of reverse transcriptase enzyme: 2U / reaction; annealing temperature: 63°C; concentration of betaine for GI: 1M, for GII: 0.8M; incubation time: 60 minutes) with detective sensitivity 10 copies/reaction and specificity of 100% for tested strains.
