Antioxidant effects of cleistocalyx operculatus in vitro and in diabetic rats

Tác dụng chống oxy hoá của nụ Vối, một nguyên liệu thông dụng dùng để chế nước uống của người Việt Nam, đã được nghiên cứu trên ống nghiệm và chuột tiểu đường. Trên ống nghiệm, bột Nụ Vối triết tách từ nụ Vối có hàm lượng polyphenol và hàm lượng flavonoid cao, đồng thời chỉ ra tác dụng chống oxy hoá mạnh khi so với bột triết tách từ lá ổi và bột chiết tách từ lá chè xanh. Trên chuột tiểu đường, sau 8 tuần cho uống nụ Vối (500 mg bột nụ Vối chiết tách/kg thể trọng) cho thấy nồng độ peroxy hoá lipid trong máu và ở mắt thấp hơn có nghĩa thống kê ở nhóm chuột uống nụ Vối so với nhóm chứng. Hơn nữa, sự phục hồi hoạt động của một số men chống oxy hoá (SOD, GST), cũng như nồng độ GSH ở gan đã được quan sát thấy trên nhóm chuột uống nụ Vối. Các kết quả đã chỉ ra hiệu quả chống oxy hoá của nụ Vối và có thể coi nụ Vối như một nguyên liệu đầy hứa hẹn trong phòng chống bệnh liên quan đến tăng stress oxy hóa.

Abstract:
The antioxidant activities and hypolipidemic effects of aqueous extract from flower buds of Cleistocalyx operculatus (COB) (Roxb.) Merry and Perry, a commonly used material for drink preparation in Vietnam, was investigated in vitro and in diabetic rats. In vitro, the aqueous extract of COB which has highest phenolic and flavonoid contents showed a strong antioxidant activity when compared to green tea and guava leaf extracts. Oral administration of aqueous extract from COB (500 mg /kg body weight/day) on streptozotocin (STZ) induced diabetic rats for 8 weeks showed significant recovery in the activities of antioxidant enzymes (SOD, GST) and GSH level in liver with a marked decrease in the lipid peroxide level in the liver and lens of the COB treated-diabetic rats. These results indicate that COB showed antioxidant effects and may be considered as a promising material for the prevention of diabetic complications and oxidative stress.
