Nấm men Saccharomyces cerevisiae đã được tối ưu hóa quá trình nhân giống trong môi trường dịch bưởi nhằm thu được mật độ tế bào cực đại. Nghiên cứu tiến hành khảo sát ảnh hưởng của nồng độ chất khô hòa tan ban đầu (độ Brix) và pH dịch bưởi cùng với tỷ lệ nấm men thuần khiết sử dụng trong quá trình nhân giống cũng như hàm lượng NaHSO3 và thời gian thanh trùng dịch bưởi trước khi nhân giống. Các thí nghiệm được thiết kế theo phương pháp bề mặt đáp ứng (Response surface methodology-RSM). Kết quả đã tìm ra giá trị tối ưu của các nhân tố là nồng độ chất khô hòa tan ban đầu của dịch bưởi (21,07 oBx), pH (4,14), tỷ lệ nấm men thuần (0,46%) kết hợp với hàm lượng NaHSO3 (109,50 mg/L) và thời gian thanh trùng (38,21 phút) cho mật độ tế bào nấm men cực đại theo mô hình là 8,89387.109 tế bào/mL sau 24 giờ nhân giống.

Abstract

The propagation of Saccharomyces cerevisiae in grapefruit juice has been optimized to obtain maximum population. This study investigated the effects of initial soluble dry concentration (oBrix) and pH of grapefruit juice with the ratio of pure yeast used in propagation as well as NaHSO3 concentration and the duration of pasteurization of grapefruit juice before reproduction. These experiments were designed by using the response surface methodology (RSM). The research results found optimal values of these factors as follows: the initial soluble dry concentration (21.07oBx), pH (4.14), the percentage of pure yeast (0.46%) associated with NaHSO3 concentration (109.50 mg/L) and pasteurization time (38.21 minutes), under which the highest yeast cell density was obtained. The predicted maximum density of yeast cell attained to 8.89387.109 cells/mL after 24 hours of propagation.
