Tóm tắt

Mục tiêu: Xây dựng quy trình phân tích kiểu gene của đa hình đơn nucleotide (SNP) LIPC rs2070895 bằng phương pháp nhân gen có mồi đặc hiệu alen (AS-PCR) trên mẫu ADN được tách từ máu toàn phần ở người từ 40-64 tuổi.

Phương pháp: Sử dụng phần mềm để thiết kế mồi đặc hiệu alen và các kỹ thuật sinh học phân tử để tối ưu hoá các cặp mồi, chu trình nhiệt thích hợp. Phương pháp giải trình tự Sanger được sử dụng để xác định kiểu gene chuẩn của đa hình đơn nucleotide này. Các thí nhiệm được thực hiện trên 16 mẫu ADN của người Việt Nam.

Kết quả: Đã xây dựng và tối ưu hóa quy trình phân tích kiểu gene của SNP LIPC rs2070895 bằng phương pháp AS-PCR với kết quả chính xác.

Kết luận: Quy trình AS-PCR để phân tích SNP LIPC rs2070895 có độ chính xác cao, cho kết quả nhanh, chi phí phù hợp và có thể lặp lại trên quần thể mẫu lớn hơn cũng như ở các quần thể khác nhau nhằm tiến tới thực hiện nghiên cứu mối liên quan của đa hình này với bệnh rối loạn lipid máu ở người Việt Nam.

Abstract

Aims: To develop a protocol for geneotyping the LIPC rs2070895 single nucleotide polymorphism (SNP), using the allele-specific primer (AS-PCR) method.

Methods: Allele-specific primers were designed and molecular biology techniques were used to optimize primer pairs and appropriate thermal cycles. Sanger sequencing method was used to determine the standard genotypes of the SNP. The experiments were applied on 16 DNA samples of Vietnamese people.

Results: A protocol for geneotyping the rs2070895 has been developed and optimized using the AS-PCR method with accurate results.

Conclusion: The AS-PCR protocol for analyzing rs2070895 polymorphism has high accuracy, fast results, low cost and it can be applied on larger sample samples in different populations to conduct research on the association of this polymorphism with dyslipidemia in Vietnamese people.
